Background:Urinary Trypsin Inhibitor (UTI), also called Ulinastatin or Bikunin, is a kind of glucoprotein with a molecular weight of about 67,000 derived from human urine. As a proteinase inhibitor, UTI inhibits the activity of trypsin, chymotrypsin, lipase, hyaluronidase, granulocyte elastase and other pancreatic enzymes. UTI is an effective for pancreatitis, hemorrhagic, traumatic , endotoxic shocks and skin shrinkage.

Description
:Recombinant urinary trypsin inhibitor (rh-UTI) produced in yeast (P. pastoris) is a single, glycosylated, polypeptide chain containing 143 amino acids, and having a molecular mass of approximately 20,000. Predicted pI=4.50.
rh-UTI is purified by proprietary chromatographic techniques, and it has higher biological activity and purity than extracted from urine.

Physical Appearance & Composition
Colorless and transparent liquid.
rh-UTI can be supplied as the lyophilized power or stored in PBS solution.


Stability
rh-UTI in solution although stable at room temperature for 4 weeks, should be stored below -20
ºC. For long term storage it is recommended to add a carrier protein (eg . 0.1% gelatin ) or be lyophilized power .
Please avoid freeze-thaw cycles.

Purity
Greater than 98.0% as determined by:
(a) Analysis by RP-HPLC.
(b) RT- FPLC.
(c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.


Amino acid sequence

The complete amino acid sequence of  rh-UTI as follows,

Dimers and aggregates
Less than 1% as determined by silver-stained SDS-PAGE gel analysis.


Specific Activity

The bio-activity of rh-UTI should be greater than 5000 U/mg (BAEE method).

Endotoxin:
Less than 0.1EU/mg of rh-UTI.

Protein content
Protein quantitation was carried out by two methods:
(a) UV spectroscopy at 280 nm.
(b) Lorry method.
According to the client’s request, the concentration of the bulk could be changed from 0.5~10mg/mL.
